Unlocking the Biology of Aging
Mojo BioPharma's Longevity Sciences division is pioneering Klotho protein therapeutics — targeting the fundamental molecular pathways that govern cellular aging, healthspan, and human longevity.
The Science of Klotho
Klotho is a naturally occurring protein that declines dramatically with age. First identified as an aging-suppressor gene in 1997, Klotho has since emerged as one of the most compelling targets in longevity biology. Mice engineered to overexpress Klotho live 20–30% longer than controls; those deficient in Klotho age rapidly and die prematurely.
Our research focuses on restoring and augmenting Klotho signaling to counteract the molecular hallmarks of aging — including cellular senescence, mitochondrial dysfunction, chronic inflammation, and epigenetic drift. We believe Klotho represents a master regulator of the aging process with therapeutic potential across multiple age-related conditions.
Mechanism of Action
Klotho functions as both a membrane-bound co-receptor and a circulating hormone. In its soluble form, Klotho modulates FGF23 signaling, suppresses oxidative stress, inhibits the insulin/IGF-1 pathway, and activates Wnt signaling — all pathways implicated in the aging process. Our therapeutic approach targets the restoration of soluble Klotho to youthful physiological levels.

Longevity Sciences Programs
MBP-101 is our lead Klotho replacement program, designed to restore circulating Klotho levels to those of young adults. Preclinical studies demonstrate improvements in metabolic function, cognitive performance, and markers of cellular aging.
- Systemic Klotho restoration
- Metabolic & cognitive endpoints
- Favorable preclinical safety profile
MBP-102 targets the role of Klotho in neuroprotection and synaptic plasticity. Emerging evidence links Klotho deficiency to accelerated cognitive decline and neurodegenerative pathology, positioning this program at the intersection of longevity and CNS medicine.
- Neuroprotective mechanism
- Synaptic plasticity targets
- Alzheimer's & cognitive aging focus
